Prepare the brownie batter according to the box directions, substituting the required water with the melted butter. Mix well.
Pour the batter into a greased 9×13 inch baking pan. Bake according to package directions for fudgy brownies, usually around 20-25 minutes at 350 degrees F. Let cool completely.
Prepare the peanut butter layer: In a medium bowl, beat together the peanut butter, powdered sugar, milk, and vanilla extract until smooth and creamy.
Spread the peanut butter mixture evenly over the cooled brownie layer. Place the pan in the refrigerator for at least 30 minutes to allow the layer to set.
